RESUMO

Spider mites Tetranychus urticae, Tetranychus macfarlanei and Oligonychus biharensis are considered to be highly polyphagous in nature and causes severe damage to a wide range of plants around the world. Amblyseius paraaerialis is an efficient phytoseiid predator of spider mites with a potential to survive on both natural and alternative diets. Evaluation of predatory potential and prey stage preference provides valuable information on the efficacy of the predatory species in controlling mite population. Feeding experiments were conducted on mulberry leaf discs under the laboratory conditions of 30 ± 2 °C and 70 ± 5% relative humidity (RH). After 24 h of feeding experiment, the adult female predator exhibited a significant preference in feeding towards the eggs of T. macfarlanei (42.6%) and the larval stages of T. urticae (46%) and O. biharensis (25.3%). The mass rearing possibilities of A. paraaerialis was tested by tracking and comparing the developmental duration of individual life stages on varied food sources like, honey, castor (Ricinus communis) pollen, honey-pollen mixture and mixed life stages of T. urticae. The predator was failed to complete its development on honey and pollen when supplied separately. However it was successfully developed on honey-pollen mixture and mixed life stages of T. urticae. The developmental studies unravelled a shortest developmental duration and an extended adult longevity and lifespan of A. paraaerialis when reared on the alternative diet, thus opened up the mass rearing possibility of the predatory species under laboratory conditions.

Egg storage duration can affect embryo mortality, hatching characteristics, hatching time, and post-hatch chick quality. In order to assess these effects, the impact of storage duration (5 days, 10 days, 15 days) and short incubation period during egg storage (SPIDES) investigated further 18, 900 eggs of broiler breeder (ROSS 308) in 3 × 2 factorial arrangement design. In the SPIDES treatment, the egg shell temperature was raised from its storage temperature (18 °C) and held at 100 °F for 3.5 h. Storage periods could significantly (P < 0.05) influence on embryo mortality (total, early, middle, and late), hatchability of both the total eggs and fertile eggs. The SPIDES treatment had a significant (P < 0.05) impact on a lower embryonic death rate and improved egg hatchability. Eggs stored for 5 days and eggs treated with SPIDES significantly (P < 0.001) shorten hatching time, batch's 90% hatching time (T 90% H), mean hatching time (MHT), maximal hatching period (MHP), and hatching window (HW). Chick quality was also determined, whereas storing eggs for 5 days and using the SPIDES treatment resulted in enhanced (P < 0.001) chick weight relative to egg weight (CW/EW), activity (AC), and chick quality score (CQS). The residual yolk sac weight (RYSW), unhealed navel (UHN %), and dirty feather (DF%) recorded the lowest (P < 0.001) values compared to long storage periods and control group. Finally, stored for 5 days treated by SPIDES positively affected the hatchability characteristics, the shortening hatching time, and the quality of chicks. Regarding the results, it was confirmed that using the SPIDES treatment to prevent the harmful effects of broiler eggs being stored for an extended period of time is a viable option.

Objective: To present an overview of different approaches and recent advances for long-term preservation of germ cells and gonadal tissues at ambient temperatures. Methods: Review of the existing literature. Results: Preserving viable spermatozoa, eggs, embryos, and gonadal tissues for the long term is critical in human fertility treatment and for the management of animal populations (livestock, biomedical models, and wild species). The need and number of banked germplasms are growing very fast in all disciplines, but current storage options at freezing temperatures are often constraining and not always sustainable. Recent research indicates that structures and functions of gametes or gonadal tissues can be preserved for the long term using different strategies based on dehydration and storage at supra-zero temperatures. However, more studies are needed in rehydration and reanimation of germplasms (including proper molecular and cellular evaluations). Conclusions: While a lot of research is still warranted to optimize drying and rehydration conditions for each sample type and each species, alternative preservation methods will change the paradigm in fertility preservation and biobanking. It will transform the way we maintain and manage precious biomaterials for the long term. Lay summary: Living sperm cells, eggs, embryos, and reproductive tissues can be preserved at freezing temperatures for human fertility treatments and used to manage breeding in livestock, laboratory animals, and wild species through assisted reproduction. These cells can be stored in cell banks and demand for them is growing fast. However, current long-term storage options at freezing temperatures are expensive. Instead of using low temperatures, recent research indicates that these cells can be dried and stored above freezing temperatures for an extended amount of time. While a lot of research is still needed to optimize how different samples are dried and rehydrated, alternative methods of preserving cells will make fertility preservation and cell banking easier. It will also transform the way we keep and manage samples for the long term.

Since they were first introduced to the United States more than 50 years ago, invasive carp have rapidly colonized rivers of the Mississippi River Basin, with detrimental effects on native aquatic species. Their continued range expansion, and potential for subsequent invasion of the Great Lakes, has led to increased concern for the susceptibility of as-yet uncompromised lotic and lentic systems in the central United States. Because invasive carp eggs and larvae must drift in the river current for the first several days following spawning, numerical drift modeling has emerged as a useful technique for determining whether certain river systems and reaches have the potential to support suspension-to-hatching survival of invasive carp eggs, a critical first step in recruitment. Here we use one such numerical modeling approach, the Fluvial Egg Drift Simulator (FluEgg), to estimate bighead carp (Hypophthalmichthys nobilis) egg hatching success and larval retention in a 47.8-kilometer (km) reach of the multi-thread St. Croix River, Minnesota and Wisconsin, United States. We explore three approaches for obtaining the hydraulic data required by FluEgg, parameterizing the model with either (a) field hydraulic data collected within the main channel during a high-flow event, or hydraulic data output from a one-dimensional hydrodynamic model with both (b) steady, and (c) unsteady flows. We find that the three approaches, along with the range of water temperatures and discharge used in simulations, produce vastly different predictions of streamwise transport and in-river egg hatching probability (0% for field data, 0 to 96% for steady-state hydraulic modeling, and 1.8 to 65% for unsteady modeling). However, all FluEgg simulations, regardless of the source of hydraulic data, predicted that no larvae reach the gas bladder inflation stage within the study reach where nursery habitat is abundant. Overall, these results indicate that the lower St. Croix River is suitable for invasive carp spawning and egg suspension until hatching for a range of discharge and water temperatures. These results highlight the role of complex channel hydraulics and morphology, particularly multi-thread reaches, and their inclusion in ecohydraulic-suitability modeling to determine susceptibility of river systems for invasive carp reproduction. Our work also emphasizes the scientific value of multi-dimensional hydrodynamic models that can capture the spatial heterogeneity of flow fields in geomorphically complex rivers. This work may help to guide management efforts based on the targeted monitoring and control and improve invasive carp egg and larvae sampling efficiency.

Gypsy moth Lymantria dispar (GM) is a polyphagous insect and one of the most significant pests in the forests of Eurasia and North America (U.S. and Canada). Accurate information on GM cold-hardiness is needed to improve methods for the prediction of population outbreaks, as well as for forecasting possible GM range displacements due to climate change. As a result of laboratory and field studies, we found that the lower lethal temperature (at which all eggs die) range from -29.0 °C to -29.9 °C for three studied populations of L. dispar asiatica, and no egg survived cooling to -29.9 °C. These limits agree, to within one degree, with the previously established cold-hardiness limits of the European subspecies L. dispar, which is also found in North America. This coincidence indicates that the lower lethal temperature of L. dispar is conservative. Thus, we found that the Siberian populations of GM inhabit an area where winter temperatures go beyond the limits of egg physiological tolerance, because temperatures often fall below -30 °C. Apparently, it is due to the flexibility of ovipositional behavior that L. dispar asiatica survives in Siberia: the lack of physiological tolerance of eggs is compensated by choosing warm biotopes for oviposition. One of the most important factors contributing to the survival of GM eggs in Siberia is the stability of the snow cover.

The vitelline layer (VL) of a sea urchin egg is an intricate meshwork of glycoproteins that intimately ensheathes the plasma membrane. During fertilization, the VL plays important roles. Firstly, the receptors for sperm reside on the VL. Secondly, following cortical granule exocytosis, the VL is elevated and transformed into the fertilization envelope (FE), owing to the assembly and crosslinking of the extruded materials. As these two crucial stages involve the VL, its alteration was expected to affect the fertilization process. In the present study, we addressed this question by mildly treating the eggs with a reducing agent, dithiothreitol (DTT). A brief pretreatment with DTT resulted in partial disruption of the VL, as judged by electron microscopy and by a novel fluorescent polyamine probe that selectively labelled the VL. The DTT-pretreated eggs did not elevate the FE but were mostly monospermic at fertilization. These eggs also manifested certain anomalies at fertilization: (i) compromised Ca2+ signaling, (ii) blocked translocation of cortical actin filaments, and (iii) impaired cleavage. Some of these phenotypic changes were reversed by restoring the DTT-exposed eggs in normal seawater prior to fertilization. Our findings suggest that the FE is not the decisive factor preventing polyspermy and that the integrity of the VL is nonetheless crucial to the egg's fertilization response.

Cell-cell fusion is limited to only a few cell types in the body of most organisms and sperm and eggs are paradigmatic in this process. The specialized cellular mechanism of fertilization includes the timely exposure of gamete-specific interaction proteins by the sperm as it approaches the egg. Bindin in sea urchin sperm is one such gamete interaction protein and it enables species-specific interaction with a homotypic egg. We recently showed that Bindin is essential for fertilization by use of Cas9 targeted gene inactivation in the sea urchin, Hemicentrotus pulcherrimus. Here we show phenotypic details of Bindin-minus sperm. Sperm lacking Bindin do not bind to nor fertilize eggs at even high concentrations, yet they otherwise have wildtype morphology and function. These features include head shape, tail length and beating frequency, an acrosomal vesicle, a nuclear fossa, and they undergo an acrosomal reaction. The only phenotypic differences between wildtype and Bindin-minus sperm identified is that Bindin-minus sperm have a slightly shorter head, likely as a result of an acrosome lacking Bindin. These data, and the observation that Bindin-minus embryos develop normally and metamorphose into normal functioning adults, support the contention that Bindin functions are limited to species-specific sperm-egg interactions. We conclude that the evolutionary divergence of Bindin is not constrained by any other biological roles.

A complex interplay of biotic and abiotic factors underpins the distribution of species and operates across different levels of biological organization and life history stages. Understanding ecosystem engineer reproductive traits is critical for comprehending and managing the biodiversity-rich habitats they create. Little is known about how the reproduction of the reef-forming worm, Sabellaria alveolata, varies across environmental gradients. By integrating broad-scale environmental data with in-situ physiological data in the form of biochemical traits, we identified and ranked the drivers of intraspecific reproductive trait variability (ITV). ITV was highest in locations with variable environmental conditions, subjected to fluctuating temperature and hydrodynamic conditions. Our trait selection pointed to poleward sites being the most physiologically stressful, with low numbers of irregularly shaped eggs suggesting potentially reduced reproductive success. Centre-range individuals allocated the most energy to reproduction, with the highest number of intermediate-sized eggs, whilst equatorward sites were the least physiologically stressful, thus confirming the warm-adapted nature of our model organism. Variation in total egg diameter and relative fecundity were influenced by a combination of environmental conditions, which changed depending on the trait and sampling period. An integrated approach involving biochemical and reproductive traits is essential for understanding macro-scale patterns in the face of anthropogenic-induced climate change across environmental and latitudinal gradients.

BACKGROUND: The diseases for which Aedes aegypti is a vector are worrisome. The high vector competence of this mosquito, as well as its anthropophilia and ability to adapt to the urban environment, allows it to exploit many habitats, making its prevention an arduous task. Despite current disease control measures focused on the mosquito, the effectiveness in containing its dispersion still requires improvement; thus greater knowledge about this insect is fundamental. METHODS: Aedes aegypti egg morphology and embryonic development were analyzed from eggs of the insectary of the Institute of Biomedical Sciences of the University of São Paulo. Optical (light and confocal) and electronic (transmission and scanning) microscopy were used to analyze the morphological and ultrastructural features of the eggs. Embryos were observed in the initial (0-20.5 h after egg-laying), intermediate (20.6-40.1 h after egg-laying), and final (40.2-61.6 h) stages of development, and kept at a temperature of 28 °C ± 1 °C until collection for processing. RESULTS: Eggs of Ae. aegypti were whitish at the time of oviposition, and then quickly became black. The egg length was 581.45 ± 39.73 µm and the width was 175.36 ± 11.59. Access to the embryo was difficult due to the egg morphology, point of embryonic development, and difficult permeability of the exochorion (mainly in fixation). Only about 5% of the collected eggs were successfully processed. In the initial stage of embryonic development, characteristics suggestive of intense cellular activity were found. In the intermediate stage, the beginning of the segmentation process was evident. In the final phase, it was possible to differentiate the cephalic region and the thoracic and abdominal segments. CONCLUSION: The chorion was found to be an important protective barrier and a limiting factor for the evaluation of the embryos and mosquito embryonic cells, indicating that further studies need to be carried out to identify the reason that this occurs.

RESUMO

Self-organization of and by the cytoskeleton is central to the biology of the cell. Since their introduction in the early 1980s, cytoplasmic extracts derived from the eggs of the African clawed-frog, Xenopus laevis, have flourished as a major experimental system to study the various facets of cytoskeleton-dependent self-organization. Over the years, the many investigations that have used these extracts uniquely benefited from their simplified cell cycle, large experimental volumes, biochemical tractability and cell-free nature. Here, we review the contributions of egg extracts to our understanding of the cytoplasmic aspects of self-organization by the microtubule and the actomyosin cytoskeletons as well as the importance of cytoskeletal filaments in organizing nuclear structure and function.

Assisted reproductive technologies (ARTs) have developed considerably in recent years; however, they cannot rectify germ cell aplasia, such as non-obstructive azoospermia (NOA) and oocyte maturation failure syndrome. In vitro gametogenesis is a promising technology to overcome infertility, particularly germ cell aplasia. Early germ cells, such as primordial germ cells, can be relatively easily derived from pluripotent stem cells (PSCs); however, further progression to post-meiotic germ cells usually requires a gonadal niche and signals from gonadal somatic cells. Here, we review the recent advances in in vitro male and female germ cell derivation from PSCs and discuss how this technique is used to understand the biological mechanism of gamete development and gain insight into its application in infertility.

Nepotism was initially theoretically predicted and sometimes found to trigger the selection of specific larvae to be reared as queens in the honeybee Apis mellifera. Although the importance of selecting the next queen for a colony indicates that it should not occur at random, nepotism is increasingly considered unlikely in eusocial insect societies. Different prenatal maternal supplies of embryos have been found to impact fitness in many other species and therefore could be a possible trigger underlying the likelihood of being raised as a queen. We offered related or unrelated larvae from six colonies originating from eggs of different weights for emergency queen rearing in queenless units with worker bees from these six colonies. We showed that nurses did not significantly prefer related larvae during queen rearing, which confirms the theory that different relatedness-driven kin preferences within a colony cannot be converted into a colony-level decision. However, we found that larvae originating from heavier eggs were significantly preferred for queen breeding. Studies on other species have shown that superior maternal supply is important for later reproductive success. However, we did observe tendencies in the expected direction (e.g., queens that hatched from heavier eggs had both more ovarioles and a shorter preoviposition period). Nevertheless, our data do not allow for a significant conclusion that the selection of larvae from heavy eggs truly offers fitness advantages.

Thermal pollution leads to short-term heat shock in aquatic invertebrates; however, the modulation of tolerance and life history of these invertebrates by thermal stress varies among regions, phenology, species, and their acclimation. To assess the effect of thermal shock, we conducted experiments on Daphnia magna and Eurytemora carolleeae at 25 °C, 30 °C, 35 °C, and 40 °C (in relation to 20 °C) in a different exposure time of the stressor (10, 30, and 60 min). The results showed that short-term heat shock leads to increased mortality and reduced fertility of the studied planktonic crustaceans. D. magna was more resistant to thermal shock than E. carolleeae according to all variants of exposure based on the calculated LT50 values for 24, 48, and 72 h. Thermal shock decreased the potential of the Daphnia population in terms of the total number of births, however, with regard to individual reproductive abilities, the non-lethal heat shock did not reduce the birth rate. Although Eurytemora is more sensitive to thermal shock than Daphnia, the type of parental care in Eurytemora might be more favorable for offspring survival following thermal shock than in Daphnia. In Eurytemora, despite maternal deaths, a relatively high number of newborns who survived high temperatures were observed. The obtained results can help to understand the ecological processes occurring due to anthropogenic thermal pollution.

Schistosomiasis is a neglected tropical disease of public health concern. The most devastating pathology in schistosomiasis japonica and mansoni is mainly attributed to the egg-induced granulomatous response and secondary fibrosis in host liver, which may lead to portal hypertension or even death of the host. Schistosome eggs induce M2 macrophages-rich granulomas and these M2 macrophages play critical roles in the maintenance of granuloma and subsequent fibrosis. Reactive oxygen species (ROS), which are highly produced by stimulated macrophages during infection and necessary for the differentiation of M2 macrophages, are massively distributed around deposited eggs in the liver. However, whether ROS are induced by schistosome eggs to subsequently promote M2 macrophage differentiation, and the possible underlying mechanisms as well, remain to be clarified during S. japonicum infection. Herein, we observed that extensive expression of ROS in the liver of S. japonicum-infected mice. Injection of ROS inhibitor in infected mice resulted in reduced hepatic granulomatous responses and fibrosis. Further investigations revealed that inhibition of ROS production in S. japonicum-infected mice reduces the differentiation of M2, accompanied by increased M1 macrophage differentiation. Finally, we proved that S. japonicum egg antigens (SEA) induce a high level of ROS production via both nicotinamide adenine dinucleotide phosphate (NADPH) oxidase 2 (NOX2) and mitochondria in macrophages. Our study may help to better understand the mechanism of schistosomiasis japonica-induced hepatic pathology and contribute to the development of potential therapeutic strategies by interfering with ROS production.

RESUMO

In vivo electroporation has become a key technique to study genetic mechanisms of brain development. However, electroporation of the embryonic pallium in oviparous species, interesting for evolutionary studies but distinct from in utero electroporation, is quite infrequent. Here, we detail the in ovo electroporation of the developing pallium in chick and snake embryos. This protocol allows gene manipulation through introducing exogenous DNA into brain progenitor cells and can be adapted to any type of gene manipulation of the embryonic telencephalon. Avian brood parasitism is costly for the host, in many cases leading to the evolution of defenses like discrimination of parasitic eggs. The parasite, in turn, may evolve mimetic eggs as a counter-adaptation to host egg rejection. Some generalist parasites have evolved host-specific races (gentes) that may mimic the eggs of their main hosts, while others have evolved 'jack-of-all-trades' egg phenotypes that mimic key features of the eggs of several different host species. The Asian koel (Eudynamys scolopaceus) is a widely distributed generalist brood parasite that exploits a wide range of host species. Based on human vision, previous studies have described Asian koel eggs as resembling those of its main host, the house crow (Corvus splendens). Using measurements of egg length and breadth, digital image analysis, reflectance spectrophotometry and avian visual modelling, we examined Asian koel egg variation and potential mimicry in egg size and shape, and eggshell pattern and color in three sympatrically occurring host species in Bangladesh: the common myna (Acridotheres tristis), house crow, and long-tailed shrike (Lanius schach). We found some differences among Asian koel eggs laid in different host nests: a) Asian koel eggs in long-tailed shrike nests were larger than those laid in common myna and house crow nests, and b) Asian koel eggs in house crow nests were less elongated than those in common myna nests. However, these changes in Asian koel egg volume and shape were in the opposite direction with respect to their corresponding host egg characteristics. Thus, our study found no evidence for Asian koel host-specific egg mimicry in three sympatrically occurring host species.

Fall armyworm (FAW; Spodoptera frugiperda), an exotic moth which recently invaded Africa, is a highly destructive pest of cereals especially maize a highly valued staple crop in Nigeria. The use of natural enemies such as predators or parasitoids for FAW control is more economically viable and environmentally safer than currently recommended synthetic insecticides. Natural enemies to combat the pest have not yet been reported in Nigeria. An exploration for the pests' natural enemies was undertaken by collecting FAW eggs and larvae from maize fields. These were reared in the laboratory for emergence, identification and efficacy as natural enemies. This yielded Euplectrus laphygmae (Hymenoptera: Eulophidae); Telenomus remus (Hymenoptera: Platygastridae) and Trombidium sp. (Acari.: Trombidiidae). Cotesia or Apanteles spp. were inferred to occur since Stictopisthus sp. (Hym.: Ichneumonidae), a secondary parasitoid, that attacks cocoons of Microgasterinae (e.g. Cotesia, Apanteles etc.) also emerged. Species of yet-to-be identified predators were also observed in various niches of maize plants. A positive relationship was found between FAW instar and the number of E. laphygmae eggs/instar ranging, on average, from 1.5 on second instar to 5.5 on fourth instars hosts. Parasitism rate of T. remus on FAW eggs was 100%. Parasitic mite infestation resulted in increasing paleness, reduced feeding, growth and movement as well as death of FAW 1st instars. Thus, the occurrence of FAW natural enemies in Nigeria calls for advocacy campaign to incorporate their use into integrated pest management strategies that attract and allow natural enemies to thrive for FAW management.

Solar ultraviolet radiation (UV) can have a wide range of negative effects on animal fitness that take place not only during, but also after exposure (carryover effects). UV-induced carryover effects and potential adaptations to avoid or mitigate them are understudied in terrestrial animals, including arthropods and their potentially most vulnerable life stages. The spined soldier bug, Podisus maculiventris, increases the emergence of its eggs that are exposed to UV radiation by coating them in sunscreen-like pigmentation, but consequences of these conditions of embryonic development for nymphs and adults are unknown. We measured stink bug nymph survival, adult size and sex ratio following exposure of differently pigmented eggs across a range of UV intensities. Nymph survival to adulthood decreased with higher intensity of embryonic UV exposure and this carryover effect decreased with higher level of egg pigmentation, similar to previously observed effects on embryonic survival. Nymph development time, adult size and sex ratio were not affected by embryonic exposure to UV radiation nor by photoprotective egg pigmentation. This study is the first to demonstrate the potential for lethal carryover effects of UV radiation in terrestrial insects, highlighting the need for more studies of how this pervasive environmental stressor can affect fitness across life stages.
